Semaglutide is a synthetic version of GLP-1 (glucagon-like peptide-1), a 30-residue gut hormone. Native GLP-1 has a half-life of about two minutes in plasma, which makes it useless for any extended experiment. Semaglutide solves that problem with a couple of amino acid substitutions and a fatty-acid side chain that lets it bind serum albumin and stick around.

Researchers studying GLP-1 receptor pharmacology have been using semaglutide in cell-model work since the late 2010s. Most of the published preclinical literature looks at receptor desensitization, biased agonism, and how GLP-1 signaling interacts with islet-cell metabolism in rats and mice.
