Nicotinamide adenine dinucleotide (NAD+) is a pyridine dinucleotide coenzyme present in all living cells, first isolated in 1906 by Arthur Harden and William John Young. As one of the most extensively characterized small molecules in biochemistry, NAD+ functions as an electron carrier in cellular metabolism and serves as a co-substrate for several classes of enzymes, including sirtuins and PARPs.

Research Context

Preclinical research has investigated NAD+ as a coenzyme in redox reactions and as a co-substrate consumed by NAD+-dependent enzymes including sirtuin deacylases, PARPs, and CD38. Laboratory studies have examined cellular NAD+ pools in models of mitochondrial bioenergetics, DNA damage response, and senescence biology.
